Skrining dan Identifikasi Molekuler Gen Penyandi 16S rRNA Bakteri Penghasil Enzim Tanase dari Hutan Lawu Utara

Abstract


Tanin merupakan senyawa fenolik bersifat rekalsitran dan toksik bagi sebagian besar mikroorganisme. Degradasi tanin oleh tanase fungi telah banyak dilaporkan, namun laporan yang berkaitan dengan kemampuan bakteri untuk mendegradasi tanin masih sedikit. Dalam aplikasinya, tanase banyak digunakan sebagai biokatalisator dalam berbagai industri. Sebagian besar biomassa di dasar hutan Lawu Utara berupa tumpukan daun pinus dengan kandungan tanin cukup tinggi, diduga memiliki potensi cukup melimpah populasi bakteri penghasil tanase. Penelitian ini bertujuan untuk mengetahui apakah di hutan Lawu Utara terdapat bakteri penghasil tanase aktivitas tinggi dan berapa besar tingkat aktivitas tanase serta bagaimana hubungan filogenetiknya. Uji kualitatif dengan mengukur indek degradasi asam tanat. Uji kuantitatif digunakan spektrofotometer UV-Vis, λ 310 nm. Identifikasi takson berdasarkan sekuen gen penyandi 16S rRNA. Terdapat 6 isolat memiliki tanase aktivitas tinggi. Aktivitas tertinggi 44,72 U/ml terdapat pada isolat B03 teridentifikasi memiliki similaritas sebesar 98% dengan genus Pseudomonas.
